ABSTRACT

Leptospirosis, a zoonotic disease that is caused by many serovars which are more than 200 in the world, is an emerging worldwide disease. Accurate and rapid diagnostic tests for leptospirosis are a critical step to diagnose the disease. There are some commercial kits available for diagnosis of leptospirosis, but the obscurity of a species- or genus-specific antigen of pathogenic Leptospira interrogans causes the reduced sensitivity and specificity. In this study, the polysaccharide derived from lipopolysaccharide (LPS) of nonpathogenic Leptospira biflexa serovar patoc was prepared, and the antigenicity was confirmed by immunoblot and enzyme linked immunosorbent assay (ELISA). The performance of the rapid diagnostic test (RDT) kit using the polysaccharide as a diagnostic antigen was evaluated in Korea, Bulgaria and Argentina. The sensitivity was 93.9%, 100%, and 81.0% and the specificity was 97.9%, 100%, and 95.4% in Korea (which is a rare region occurring with 2 serovars mostly), Bulgaria (epidemic region with 3 serovars chiefly) and Argentina (endemic region with 19 serovars mainly) respectively. These results indicate that this RDT is applicable for global diagnosis of leptospirosis. This rapid and effective diagnosis will be helpful for diagnosis and manage of leptospirosis to use and the polysaccharide of Leptospira may be called as genus specific antigen for diagnosis.

BACKGROUND: Few studies evaluated the association between nutritional disorders, quality of life and weight loss in patients undergoing bariatric surgery. AIM: To identify nutritional changes in patients undergoing bariatric surgery and correlate them with weight loss, control of comorbidities and quality of life. METHOD: A prospective cohort, analytical and descriptive study involving 59 patients undergoing bariatric surgery was done. Data were collected preoperatively at three and six months postoperatively, evaluating nutritional aspects and outcomes using BAROS questionnaire. The data had a confidence interval of 95%. RESULTS: The majority of patients was composed of women, 47 (79.7%), with 55.9% of the series with BMI between 40 to 49.9 kg/m². In the sixth month after surgery scores of quality of life were significantly higher than preoperatively (p<0.05) and 27 (67.5 %) patients had comorbidities resolved, 48 (81.3 %) presented BAROS scores of very good or excellent. After three and six months of surgery 16 and 23 presented some nutritional disorder, respectively. There was no relationship between the loss of excess weight and quality of life among patients with or without nutritional disorders. CONCLUSION: Nutritional disorders are uncommon in the early postoperative period and, when present, have little or no influence on quality of life and loss of excess weight. .


RACIONAL: Poucos estudos avaliam a associação entre distúrbios nutricionais, qualidade de vida e perda de peso em pacientes submetidos à cirurgia bariátrica. OBJETIVO: Identificar alterações nutricionais em pacientes submetidos à cirurgia bariátrica e correlacioná-las com perda de peso, controle de comorbidades e qualidade de vida. MÉTODO: Estudo de coorte, prospectivo, analítico e descritivo envolvendo 59 pacientes submetidos à cirurgia bariátrica. Os dados foram coletados no pré-operatório e aos três e seis meses pós- operatórios, quantificando aspectos nutricionais e utilizando o Bariatric Analysis and Reporting Outcomes System (BAROS) como ferramenta de sucesso. Os dados usaram intervalo de confiança de 95%. RESULTADOS: O total de mulheres foi 47 (79,7%), sendo 55,9% com IMC entre 40-49,9 kg/m². No sexto mês depois da operação os escores de qualidade de vida foram significativamente maiores do que no pré-operatório (p<0,05) e 27 (67,5%) pacientes tinham todas comorbidades resolvidas, 48 (81,3%) apresentaram conceito BAROS muito bom ou excelente. Após três e seis meses 16 e 23 pacientes apresentaram algum distúrbio nutricional, respectivamente. Não houve relação entre a perda do excesso de peso e qualidade de vida entre pacientes com ou sem distúrbio nutricional. CONCLUSÃO: os distúrbios nutricionais são pouco frequentes no pós-operatório precoce e, quando presentes, têm pouca ou nenhuma influência na qualidade de vida e na perda do excesso de peso. .

ABSTRACT

The present study aims to isolate the polysaccharides of the oleo-gum-resins of Boswellia carteri and Commiphora myrrha Family Burseraceae, identify their components and test their immuno-modulatory activity for the first time. The polysaccharides were isolated from the oleo-gum-resins of Boswellia carteri and Commiphora myrrha using different chemical and chromatographic techniques. The components of the polysaccharide hydrolysates are estimated qualitatively and quantitatively by using PC and HPLC analysis. The immuno-modulatory activity is tested by splenocyte and mitogen induced lymphocyte proliferation methods. The percentage yield of the isolated polysaccharide was 15.4% and 15.7% from Boswellia carteri and Commiphora myrrha oleo-gum-resins respectively. Galactose and ribose are the major constituting units of the polysaccharide isolated from Boswellia carteri oleo-gum-resin whereas ribose and galacturonic acid are the major constituting units of the polysaccharide isolated from Commiphora myrrha oleo-gum-resin. The polysaccharide of B. carteri oleo-gum-resin showed mild immuno-stimulatory activity at a dose of 5microg/ml, while that of C. myrrha oleo-gum-resin showed no activity at this dose. Chromatographic investigation of the polysaccharides isolated from Boswellia carteri and Commiphora myrrha oleo-gum-resins by PC and HPLC analysis demonstrated their qualitative and quantitative variation in their chemical composition. Higher concentrations of the polysaccharides could be tested for immuno-modulatory activity

ABSTRACT

We evaluated the functional activity of Haemophilus influenzae B (Hib) antibodies elicited in a group of infants immunized with the diphtheria-tetanus-pertussis vaccine combined with an Hib vaccine produced totally in Brazil after technological transfer of Hib vaccine production from Glaxo SmithKline, Belgium. Blood samples from immunized infants (N = 985) were collected for the determination of Hib antibodies. Total Ig and IgM and IgG subclasses of antibodies against polyribosyl ribitol phosphate (PRP) were analyzed by ELISA. Almost all vaccinees (97.56 percent, 961/985) developed a strong anti-PRP IgG antibody response (¡Ý1.0 ¦Ìg/mL), while an anti-PRP IgM response was observed in 64.24 percent (634/985) of them (¡Ý0.15 ¦Ìg/mL). Only 18.88 percent (186/985) of the infants in the group with high PRP antibody IgG concentrations (¡Ý1.0 ¦Ìg/mL) developed a high IgM antibody response. Anti-PRP IgG antibody levels were significantly higher than anti-PRP IgM. These results demonstrate the predominance of IgG antibodies over IgM antibodies in response to PRP, with a ratio of 17:1. IgG antibodies were predominantly of the IgG1 subclass. An increase in IgG avidity was also observed during the course of immunization.

ABSTRACT

This study evaluated the vaccination response to Haemophilus influenzae type b (Hib) in malnourished pregnant women (MN), cord blood (CB) and in infants at two and six months of age for comparison with a control group (C). Twenty-eight malnourished pregnant women and 29 pregnant controls were immunized with conjugated Act-HIB® in the third trimester of pregnancy. Blood samples were collected from all before the immunization, during labor (post immunization), and from CB. All infants were immunized with Hib vaccine according to normal vaccine schedule and sera were collected at two and six months of age. Antibody levels to polyribosylribitol phosphate (PRP) were similar for both groups. Preimmunization: MN 1.94 µg/mL, C 1.68 µg/mL; post-vaccination: MN 18.53 µg/mL and C 17.55 µg/mL; in CB from MN 14.46 µg/mL and from C 17.04 µg/mL. Infants from MN and C mothers presented respectively at two months: 5.18 µg/mL and 8.60 µg/mL and at six months: MN 3.42 µg/mL and C 2.18 µg/mL. Antibody levels were similar in both groups studied (p = 0.485), however the vertical transmission rate was 14 percent lower in the MN pregnant group. Levels of antibodies > 0.15 µg/mL were found in all newborns from the MN pregnant group. Pregnant MN presented an immunological response to Hib vaccine similar to group C, however, vertical transmission rate of antibodies to PRP in the MN pregnant group was 14 percent lower than that in C, suggesting a less efficient passage of antibodies within this group.

ABSTRACT

Recent evidence indicates that protein-glycan interactions play a critical role in different events associated with the physiology of T-cell responses including thymocyte maturation, T-cell activation, lymphocyte migration and T-cell apoptosis. Glycans decorating T-cell surface glycoproteins can modulate T-cell physiology by specifically interacting with endogenous lectins including selectins and galectins. These endogenous lectins are capable of recognizing sugar structures localized on T-cell surface glycoproteins and trigger different signal transduction pathways leading to differentiation, proliferation, cell cycle regulation or apoptosis. Protein-carbohydrate interactions may be controlled at different levels, including regulated expression of lectins during T-cell maturation and differentiation and the spatio-temporal regulation of glycosyltransferases and glycosidases, which create and modify sugar structures present in T-cell surface glycoproteins. This article briefly reviews the mechanisms by which protein-carbohydrate interactions modulate immunological processes such as T-cell activation, migration and apoptosis.


Las interacciones entre proteínas y glicanos juegan un papel fundamental en numerosos eventos de la regulación de la fisiología del sistema inmune, como maduración tímica, activación, migración y apoptosis de células T. Los carbohidratos son capaces de modular la fisiología linfocitaria a través de la interacción específica con lectinas endógenas como selectinas y galectinas. Estas lectinas endógenas son capaces de reconocer estructuras sacarídicas localizadas en glicoproteínas de la superficie celular y regular procesos tan diversos como proliferación, diferenciación y ciclo celular. Existen diversos niveles de control de la interacción entre lectinas y azúcares; en primer lugar podemos mencionar la expresión regulada de estas lectinas durante el desarrollo de una respuesta inmune, y en segundo lugar la regulación espacio-temporal de la actividad de glicosiltranferasas y glicosidasas cuya función es crear y modificar los azúcares específicos para estas lectinas. Existen evidencias de que la expresión y actividad de estas enzimas se regulan en forma positiva o negativa durante diferentes eventos del desarrollo, ejecución y finalización de la respuesta inmune. En este artículo se analizarán los mecanismos a través de los cuales las interacciones entre lectinas con sus carbohidratos específicos modulan en forma específica diversos procesos fisiológicos, como maduración de timocitos, migración linfocitaria, activación y diferenciación de células T y apoptosis.

ABSTRACT

An enzyme-linked immunosorbent assay was standardized for the detection of cryptococcal antigen in serum and cerebrospinal fluid. The system was evaluated in clinical samples from patients infected by human immunodeficiency virus with and without previous cryptococcosis diagnosis. The evaluated system is highly sensitive and specific, and when it was compared with latex agglutination there were not significant differences. A standard curve with purified Cryptococcus neoformans antigen was settled down for the antigen quantification in positive samples

ABSTRACT

O estudo envolve a comparaçäo entre o antigeno polissacaridico de Histoplasma capsulatum com a histoplasmina classica em inquerito epidemiologico, atraves de provas intradermicas de hipersensibilidade do tipo tardio, realizado em 115 individuos da regiäo de Santo Amaro. Os resultados revelaram 46 por cento de provas positivas com a histoplasmina classica e 51,30 por cento de resultados positivos com o antigeno polissacaridico em sua maior concentraçäo. A principal conclusao da pesquisa: e possível utilizar o antigeno polissacaridico como histoplasmina, em substituiçäo ao antigeno filtrado

Candidina, constituida de suspensao de celulas leveduriformes mortas, e comumente usada em provas intradermicas de hipersensibilidade retardada, principalmente na avaliacao da competencia imunologica do paciente, quando usada conjuntamente com outras provas intradermicas do mesmo tipo. considerando-se o estudo histopatologico de reacao positiva com este tipo de antigeno, e possivel obter uma reacao positiva nao especifica na leitura da prova intradermica. ABSTRACT

En tres voluntarios sanos se les indujo un modelo de gingivitis en la hemiarcada superior derecha (14 días sin ninguna medida de higiene bucal); la hemiarcada superior izquierda sirvió como control. Se tuvo el propósito de demostrar los patrones de expresión de diferentes residuos glicosídicos usando un grupo de ocho lectinas biotiniladas en encía sana y enferma. Se tomaron biopsias por congelación en los días 0, 7 y 14 del experimento y se procesaron con la técnica histoquímica de la inmunoperoxidasa. Fue posible determinar la adhesión selectiva de las lectinas a las diferentes estructuras de los tejidos epitelial y conectivo de la encía y se observó un cambio en la expresión de residuos sacáridos a medida que avanzaba la inflamación experimental, posiblemente atribuible a la creación de criptoepítopes mediante el clivaje del azúcar terminal de los glicanos, a la posible acción de una transferasa y a la especificidad de las lectinas por el grupo sanguíneo del paciente al que pertenecía el tejido bajo examen

Fueron examinadas las muestras de suero de 242 personas, HIV positivas, para determinar la presencia de antigeno capsular del C. neoformans, 193 de estos pacientes tenian recuentos de celulas CD4+inferiores a los 300/µl y 49 pacientes presentaron recientos superiores a este limite. Ninguno de los enfermos tenia sintomatologia que hiciese sospechar criptococosis. El antigeno capsular del C. neoformans fue determinado por uma tecnica de aglutinacion de particulas de latex previo tratamiento con pronasa (IMM, latex-Crypto antigen detection system, Immunomycologics, Oh, USA) y 61 por cento de las muestras fueron tambien examinadas mediante la tecnica de ELISA (Premier, Cryptococcal Antigen, Medirian Diagnostic INC, Cincinatti, OH, USA).

Dois antissoros produzidos em carneiros, um dos quais contra a fracao polissacaridica (Po) e outro contra componentes proteicos (Pt) de vermes adultos de Schistosoma mansoni, foram avaliados quanto ao seu desempenho na deteccao de antigenos parasitarios circulantes em pacientes com diferentes formas ciclicas da esquistossomose mansonica, atraves de um ensaio imunoenzimatico, (ELISA). O primeiro antissoro revelava antigenos parasitarios no granuloma hepatico e o segundo, nos glomerulos renais de pacientes com esquistossomose mansonica, bem como em camundongos experimentalmente infectados pelo S. mansoni....
